CML Quest-BPA
(BCR/abl with Break Point Analysis)
Break Point Analysis is assessment of transcripts corresponding to Major, Minor and Micro breakpoints. Transcripts p210, p190 & p230 are evaluated in a PCR reaction.
Breakpoint analysis is a deep incite in to Philadelphia Hybrid variants for better patient management that adds significant prognostic information about the disease.
Presence of break point corresponding transcript p190 renders the disease as aggressive & is present in 5-7% of CML patients, where as break point corresponding to transcript p210 is comparatively less aggressive & is present in 95% of CML patients.
Breakpoint corresponding to transcript p230 is found in less than 1-2% of CML patients & the same breakpoint is indolent in nature.
Methodology:
Patients DNA would be isolated, purified and subjected to PCR amplification using primers & Taqman probes. An additional amplification directed at GAPDH would be performed as a control for sample DNA quality.
